Study and detection of ERG11 gene mutation in resistant - drug Candida albicans and relation with Iranian women infertility
Authors
Abstract:
Aim and Background: Candida Albicans is the most important cause of vulvovaginal candidiasis and resistance to azoles can occur via various mechanisms including, change in the ERG11 gene. This study aim was to identification of ERG11 gene mutations in drug resistant Candida albicans isolated from patients with Candida vaginitis and its association with infertility in Iranian women. Material and methods: From 100 patients with vaginitis Candida Albicans species were isolated by using chromogen agar culture . The drug sensitivity was determined with standard protocol, diffusion disc test. Then extraction genome from resistance species and was used PCR and sequencing for investigation of ERG11 gene mutations. For detection of sperm parameters disorders and relation with infertility, normal sperm was placed adjacent to sensitive and resistance Candida Albicans by different concentrations and times. Results: From 100 patients, 40 samples of Candida albicans were isolated that 2 samples were fluconazole and ketoconazole-resistant. ERG11 gene mutations e in 2 samples were, conversion and insertion. In 10000 and 100000 CFU/ml concentrations, 4 hours and with 3 repeated, was observed significant relation to agglutination and reduction in motility of the sperms (P<0.05). Conclusion: This study results showed that antibiotic resistance can be increased in Candida albicans with mutation in the ERG11 gene and this Candida vaginitis infections, can associated with infertility in women.

Quantitation of ergosterol content and gene expression profile of ERG11 gene in fluconazole-resistant Candida albicans
Background and Purpose: The frequency of opportunistic fungal infections in immunocompromised patients, especially by Candida species, has sharply increased in the last few decades. The objective of this study was to analyse the ergosterol content and gene expression profiling of clinical isolates of fluconazole resistant Candida albicans. Materials and Methods: Sixty clinical samples were ide...
full textMUTATION IN HOTSPOT REGIONS OF THE ERG11 GENE AND FLUCONAZOLE RESISTANCE IN CLINICAL ISOLATES OF CANDIDA ALBICANS IN RASHT CITY
Background & Aims: Nowadays, the common use of azoles has led to increased resistance to azole among Candida albicans strains. Amino acid substitutions in azole target enzyme, ERG11p, is attributed to azole resistance in some clinical strains of Candida albicans. The aim of this study was to evaluate ERG11 gene mutations in fluconazole-resistant isolates of Candida albicans in Rasht. Materials...
full textEffect of biogenic selenium nanoparticles on ERG11 and CDR1 gene expression in both fluconazole-resistant and -susceptible Candida albicans isolates
Background and Purpose: Candida albicans is the most common Candida species (spp.) isolated from fungal infections. Azole resistance in Candida species has been considerably increased in the last decades. Given the toxicity of the antimicrobial drugs, resistance to antifungal agents, and drug interactions, the identification of new antifungal agents seems essential. In this study, we assessed t...

Patients with impaired immunity are at particular risk of infections with Candida albicans. Antifungal drugs such as azoles commonly used for candidiasis treatment, but drug resistance is one of the most common problems for public health. The aim of this study was to evaluate the expression of lanosterol 14-demethylase (ERG11) gene for three-drug combinations in C. albicans. Disk diffusion and ...

full textNucleotide substitutions in the Candida albicans ERG11 gene of azole-susceptible and azole-resistant clinical isolates.
One of the mechanisms of Candida albicans resistance to azole drugs used in antifungal therapy relies on increased expression and presence of point mutations in the ERG11 gene that encodes sterol 14α demethylase (14DM), an enzyme which is the primary target for the azole class of antifungals. The aim of the study was to analyze nucleotide substitutions in the Candida albicans ERG11 gene of azol...
